چکیده

Macrophages isolated from the peritoneal cavity of untreated mice and maintained in tissue culture synthesize and release prostaglandins when challenged with zymosan. These cells also selectively release lysosomal acid hydrolases under the same conditions. The major prostaglandins released into the media are found to be prostaglandins E1, E2 and 6-oxoprostaglandin F1a, whereas prostaglandin F2a is not detected. Macrophages isolated from mice that have received an intraperitoneal injection of thioglycollate broth are far less responsive to zymosan challenge. These cells require 300 microgram of zymosan to synthesize and release one-third the amount of prostaglandins released from non-stimulated macrophages exposed to 50 microgram of zymosan. In addition, thioglycollate-stimulated macrophages release less than 10% of their lysosomal acid hydrolases when exposed to 300 microgram of zymosan whereas non-stimulated cells release approximately 50% of these enzymes after treatment with 50 microgram of zymosan. The zymosan-stimulated synthesis and release of prostaglandins are completely inhibited by indomethacin, whereas the increased selective release of lysosomal acid hydrolases is not affected. Macrophages, unlike fibroblasts, do not synthesize and release prostaglandins when exposed to serum or to bradykinin.

Effect of Ganoderma Lucidum on Cytokine Release by Peritoneal Macrophages

Background: The water-soluble extract of Ganoderma lucidum (Reishi) has been used as an immunomodulator to stimulate spleen cells proliferation and cytokine expression. Objective: To investigate the effect of Ganoderma lucidum (G. lucidum) on cytokine production by mice peritoneal macrophages. Methods: Mice peritoneal macrophages were prepared by intra-peritoneal injection of 5 ml cold PBS. Per...

FIVE ALPHA DIHYDROTESTOSTERONE (5α-DHT) MAY MODULATE NITRIC OXIDE RELEASE VIA ENDOGENOU S CYTOKINES IN PERITONEAL MA CROPHA GES OF NZB/BALBc MICE

Recent studies have established that sex hormones directly or indirectly affect T and B cells and macrophages by manipulating the production of cytokines. In this study the possibility of the effect of 5a-DHT on macrophage (MΦ) nitric oxide (NO) release via interleukin-l, 6 (lL-1β, IL-6) or tumor necrosis factor-a (TNFα) was investigated. The endogenous cytokines IL-1β, IL-6 and TNF-α were ...

Selective release of lysosomal hydrolases from phagocytic cells by cytochalasin B.

1. Cytochalasin B (10mug/ml) enhances the release of rabbit polymorphonuclear leucocyte lysosomal acid hydrolases induced by retinol (vitamin A alcohol). 2. This effect is seen at doses of the vitamin that cause selective release of acid hydrolases and those causing more general enzyme release indicated by the loss of lactate dehydrogenase. 3. Cytochalasin B (2-50mug/ml) has no effect on the re...

EFFECT OF FIVE ALPHA DIHYDROTESTOSTERONE (5α-DHT) ON CYTOKINE PRODUCTION BY PERITONEAL MACROPHAGES OF NZB/BALBc MICE

One of the mechanisms involved in the regulation of the immune system by steroid hormones could be the monocytic-macrophage system. In this study the effect of the male hormone 5a-DHT on cytokine release by peritoneal macrophages (mΦ) of male and female NZB/BALBc mice was investigated. Macrophages from male mice activated with LPS produced a greater amount of IL-1β (21.8%) (p<0.05) and IL-...

Changes in Cellular Enzyme Levels and Extracellular Release of Lysosomal Acid Hydrolases in Macrophages Exposed to Group a Streptococcal Cell Wall Substance

Mouse peritoneal macrophages exposed to type-specific polysaccharide and peptidoglycan (PPG) from group A streptococci undergo marked morphologic and biochemical changes. The cells show increases in size and an increased number of lysosomes as demonstrated by vital staining with acridine orange. There are significant elevations in the levels of both lysosomal and nonlysosomal enzymes. Higher do...
